Its an application called Rainmeter, and that's just one of its many skins (which you have to search individually).
I'm sure a few mins. w/ Google will get you what you need.

Do you do Dynamic lines? or do you manually sharpen the lines of stroked lines?

Yo SM. I normally manually sharpen stroked lines for most of the pic, but for some parts like the face, I use dynamic lines.

It's not exactly my art, it's Ken Akamatsu's. I just trace from the manga then colour it. ^^
Pointers? Dunno, I haven't been doing this for a long while yet. Try and find the AQS forums and look for the colourist's thread. They can give ya lots of help.
