This is a nice theme, a lot of ideas come to mind for something like this, did a nice job of expressing yours.

Fantastic way to convey how the "irritated" is trying to maintain a steady and continuous pace of walking, inspite the aching frequency of poking he is being subordinated to on behalf of the "irritator" . I get a special kick of laugh and jiggles from the way the lil' purple fella is jumping-along frevorously, with such joy exsuding from his face.
I think the length of the comic episode is optimal, it last long enough to show us what is going on, and is being cut and reversed, just in the right time, for us to get a sense of repetitive monotony, that makes the viewer sense some of what the "irritated" is going thru mentally.
Perhaps in the future, this entire concept might get a rendition, where you would expand the story line of the animation, by introducing a dramatic break where the red one takes a moment to face the audience and make some kind of a facial expression, whether it be despair, or steaming in rage, or threatning the purple agitator... and then continuing-on with their established little routine, as if it were a symbiosis of sorts, to which they have both become accustomed to.
I'm all about the concept of pesterns and pushing people's buttons, after all... it's the premise of mi avitor.
Who would have thought that you still haven't done one so very obvious, and so very humorous emote, that not only is not on the DA list, but I have not seen it done before either. I guess it's a good sign however... prooving that emoticonists will never run out of new, fresh and innovative ideas. Excellent work Matt.

I like what you've done, its a nice little emote, but, I feel as if you could of added more.
Have you ever seen a little kid in the supermarket with his mother? (i'm sure you have) But, he gentlely taps his mother on the shoulder or whatnot, than much faster, than he begans to get hyper because his mother does not answer....etc.
I'd suggest adding more frames, have him getting continously more pessy, after a few rounds of tapping, he may began to jump, yell, or whichwhat, than have the elder emote smack him a new one.
I like your emote as it is, but extra detail never hurts
