That beautiful, wonderful line is a great culmination of how much they've grown closer together as time went on. Their relationship has undergone a gradual evolution in the show. There were always hints of love and motherly/sisterly care from the beginning, but they were mostly kept in the background as the show emphasized Spike's "no. 1 assistant" status more (not to mention the fact that Spike wasn't very well treated by staff-writers in the past).
But now? Now we have lines like this, and even more familial moments between the two in recent seasons.
- Giving them family traditions in "Hearthbreakers."
- Twilight's instinctual protection of Spike in "What About Discord" and, to a lesser extent, "A Flurry of Emotions."
- A cook-off just for fun in "A Health of Information."
- And there's probably others that don't immediately come to mind.
Their relationship is just so subtle, complex, and amazing! It's not perfect, but no relationship is truly perfect. Everyone has faults they need to learn to overcome. Thankfully, Twilight and Spike are there to help each other grow. Here's to more glorious Twi-Spike moments in Season 8!

I can totally see the points you brang up and It really does bring a tear to my eye as well. These moments truly are special in every way.
I too hope for more moments like these and maybe even Spike accidently calling Twilight his mom... perhaps it could happen in a TwiSpike bonding episode?

These moments are incredibly special, and I love them so much 
Spike calling Twilight mom would be wonderful (and the feels would be awesome!). Believe it or not, I actually like the show keeping it ambiguous and letting the audience figure out what their relationship is. But that's because I'm a fan of subtle storytelling where the audience fills in the gaps.
Me personally? I like to see them as mother-son and brother-sister, kinda like Lilo and Nani in Lilo and Stitch. I mean the show pretty much confirmed they're family already in "Once Upon a Zeppelin."
But yeah, I'd pay all the money in the world for a Twi-Spike bonding episode...preferably an travel episode because adventure stories are my favorites!
