This is adorable. I never thought of them as mother and son, but it fits, I guess. He obviously would have imprinted on her, as she was there when he hatched. Actually, this makes a lot more sense than my headcanon where they're more of a brother and sister.

Awww... that's a very sweet comic. And very early too. But as far as I'm concerned, he probably consider her more as a big sister than a mother. However, the way he showed affection toward her surely give an idea that he may consider more than a big sister feeling reinforced by Twilight motherly attitude toward Spike. But, big sisters can do as well.
But I'll just leave this at that. Maybe one day he will meet his real mother.
Twilight: Did you classified the scrolls like I asked you?
Spike: Uh... no?
T: Cleaned the bath after your long bath?
S: Uh... I was going to...
T: And the windows?
S: Twilight... I-I was busy writing this for you...
T: I know. That's okay.
S: Phew.
