Damn Eddy, five kids! Anyway, who am I to judge if he wants to bang Lee non-stop? I mostly want to comment on the designs. This is something I don't find in the Ed Edd n Eddy fan art community. Rarely do the fans ever stick to the original style. You make great use of it here by making a whole cast of unique characters that are easily distinguishable from one another. I really look up to you for that.

A Nana Mouskouri lookalike.
Next to the giant harp sitting in the corner of Eddy's brother's bedroom, there's a simple poster advertising a live show of Nana Mouskouri. I first thought he just had a very backwards taste in music, but not far from these two items there's a calender that says "dirt rock", which I'd say is more character-appropriate.
Having that said, he might just love the woman for the woman and maybe used to play along with her tunes on his harp. In his little Mouskouri corner. It made sense to me that someone like her would be his dream girl. And it makes for a funny couple.

Both sets of sisters aren't really twins, they just dress alike.
There are more characters here that look like other characters, but I didn't want to reserve certain features to just one person. That's not realistic.

Pretty good overall, but I think you might want to go back to the drawing board for Eddy and Lee's kids, they just feel really off(I think I remember an earlier version you did looked better)
Also Lee's paternal grandmother reminds me a lot of Jimmy, which is now got me imagining Lee and Jimmy being 2nd or 3rd cousins

Guess you're right, I remembered there being more differences than in reality, might have gotten it mixed up with how Version 1 had a more expanded Edd/Marie family(overall Version 1 kinda feels like it would work better than Version 2, especially feel that splitting Penelope into Bunny & Chick wasn't a good idea, they feel a lot more shallow than Penelope was, at least going off the brief snippets you wrote for each character, same for Sheldon, also overall feels like you picked the least interesting of the Edd/Marie children to survive into V2 both visually and personality wise, will admit that Casey and Penny are pretty good additions to the V2 Eddy/Lee family)
Of course this is your art and concept, so you're free to disregard my commentary if you wish, although I'm also up for discussing this topic further(I've probably missed some things while typing this up for example)

Ah, so I see.
Version 1 was made for the genderbent Ed, Edd n Eddy, though I didn't bother to change their genders, and version 2 was always meant for the regular cast. So what we see here is what I intended back in 2013.
Penelope, who is Bunny here, received no changes at all, nor did Sheldon, really. The insertion of Chick only added that Sheldon "comically" prefers Chick over Bunny, despite both of them acting the same way towards him. And that's mostly it.
I went with Marian, because she is the best combination of both her parents. It was either going to be her or the eldest from version 1; but I didn't want to go with a complete Edd clone as their only daughter. I fear version 1-Mildred is too boring without her sisters, and Martha and Marjorie have the type of personality that would only make sense when they're part of a bigger family as well. How could someone like Edd and Marie have an only child that is either dumb or a diva. These girls only have these characteristics to differentiate themselves from their other sisters. I didn't want them to be clones or be even remotely similar to each other.
Marcy is a witty prankster, who would've been a nice alternate pick simply because of the dialogue she'd deliver, but she comes closer to Eddy than Edd and Marie, so she also wasn't quite the child I wanted to present as the ultimate combination of her parents.
Marian has a friendly smile, friendly voice, but destroys people when her father isn't looking; it's mob boss Marie hiding under Edd's approachability. To me, she's the only one who can pull off the role of an only child and have her personality make sense.

Ah, yes, having re-watched the scene, he says "No, Mommy, no Auntie for Ed!" So it was his mother's arm dragging him off too his aunt. Thanks for clearing that up!

I'm sure her arm was never portrayed, but this is what I found about Ed's aunt on the Ed, Edd n Eddy Wiki:
"Ed's Aunt is the aunt of Ed and Sarah. It is unknown if she is the sister or sister-in-law of their mother or their father.
Ed would have to live with her if he received bad grades in school (it is unknown for how long), as mentioned in "Mission Ed-Possible." It is most likely that she is the sister of Ed's mom, since that's the parent shown wanting to send Ed to his aunt at the end of the episode. She has a cat which makes Ed sneeze, meaning it could be one of his allergies. She never butters Ed's toast, possibly because she has a bad relationship with him, or she simply doesn't like buttered toast. She smells like cabbage, has a mustache, and she possibly lives far away. Because she forces Ed to use toothpicks, she may have an obsession with cleanliness, although she doesn't sound very hygienic in any other part of her description."
I can't say if there were any other mentions in the show about her personality or appearance, I have to trust the fans that maintain the Wiki to have picked up every bit of information available.
