I can see why. You see in the manga, Ai introduces Ren as her "Kyoodai" which translates to sibling, which could mean either gender.
However the kanji for "Kyoudai" suggests a male sibling being present. That being said Hino-sensei sneakily didn't use kanji to write that word, which means that she left it ambiguous on purpose XD
That being said, sometimes Ren uses phrases or expressions mostly used by males in the original version, so that is a hint
In the Japanese copy I have, he started laughing cause when you put Ren and Ai's names together it becomes "RenAi" which means "Romantic love" XD
