I design/modify my frames in Photoshop, save each of them individually, then I build my animations in Ulead Gif Animator 5. You can also build them using Photoshop, but in my humble opinion Photoshop sucks at animations, it adds way too much noise when you convert the images from jpeg/png to gif.
