Mephisto reveals to Dr. Strange that he has seen a vision... of his downfall. And he kept seeing it so many times, and it all leads to Spider-Man being the downfall of his reign, and the more he sees the vision, it changes... to the reason he sold Peter's and MJ's marriage to him... He sees the daughter of Peter Parker and Mary Jane Waston... a Spider-Girl, to be the ultimate downfall of his reign.

My guess is... To prevent that vision to come to pass, Mephisto took Peter and MJ's marriage so Aunt May will be alive, everyone forget he's Spider-Man and their daughter won't exist to stop him in the future.

All this... cause he's scared of their future daughter that was suppose to come.

So Mephisto really was scared of Spider-Girl.
