To my knowledge, the tilde isn't used much in the English language. The only 'official' use I know of is to represent an approximation of a number, like if something is 'about 6', you can use '~6'. In mathematics I've seen it used for a similar purpose. A tilde on top of an equal sign represents that something is 'roughly equal to' something else.

I don't know why I use it as punctuation, but it's just something I enjoy doing. It's kind of a personal signature in my writing, something I put in place of a period at the end of a paragraph when I'm being informal and generally in a good mood.

On an American/English keyboard, it's on the top-left corner, to the left of the '1' key.
