I use photoshop to draw and animate these.
Each movement should be drawn on a new layer eg open mouth is one layer closed is another.

I use paint tool sai, then I move the file onto adobe Photoshop CS6 to animate~

with pixels there isn't really, sometimes making the canvas bigger itself solves the problem

well other brushes that arn't the binary tool will make it smooth, but smooth pixel art with the binary tool.. I'm not so sure
