The red one actually rides the Excess Express during Mario's trips. The Red conductor not only offers assists passengers on the ride- he's also what one would consider a Blanket PI.

One blue conductor handles tickets in Rogueport, while another handles the checkpoint near Riverside Station. The first tries to apprehend the fake Zip Toad, but the imposter shakes off the disguise, scaring the blue dude. This gives the fake time to escape, yet blue no time to say anything unexpected or profound.

Green...green...just green. (Yeah, not seeing him from skimming the footage or on Mariowiki. My mind's blank on the Green Conductor, considering Spriter's Resource shows the green head.)
