I quite like the theory of it being a kind of ranking system. Maybe there were a lot of humans who came before her, and some of them only got so far.
For instance, maybe out of the 53 that made it to the land of the corgis, 4 of them didn't make it to the exit. So, when Tulip opened the door, her rank increased (notice that we don't see her number after she opens the door, so that could potentially be the cause for it changing).

Also, wouldn't it be interesting if there was another human on the train who had the number 1 on their hand? I'm not sure what the implications of that would be, but it would be quite interesting to find out (;

That's a pretty solid theory. But there's just this one thing. The corgi land isn't that challenging of a place to get across so 4 people who couldn't make it doesn't make much sense. Unless those 4 are stuck somewhere in the previous cars but you'd think tulip would've notice them by now. Anyway that's justa minor nitpick of the theory.

Well, we don't know for sure what could've happened to any other humans. They probably didn't get stuck, but maybe they encountered the steward as well, or something similar. Not everyone would be as lucky as Tulip, considering the steward just up and left for some mysterious reason.
Also, we don't really know how long this train has been running. Maybe it's just been one human at a time, and each of them have died off before reaching the end (if there is one, considering the name of the cartoon haha)

I don't think the steward is out trying to kill the people there. It seemed as if she just want people to "return to their seats." Also another thing, the steward was busy fixing the water waaay over on the other side of the car leaving the door right there for people to go through. So i don't think the steward would be the one to stop them if they came in. 
That being said, the thing that came through the hole in the wall that broke the water, now THAT could be the thing to support the theory. Only problem is that we don't know what broke the water in the first place

True. All I know is, there could've been a number of different reasons why a few people didn't make it past that particular car.
I guess we'll just have to wait until a series is made to find out ^-^
