It really works as an homage and an inversion:
Initially Rita was the anti-Harley. She just wanted to overthrow Zedd not love him. So her seductive appearance is here is warranted. Like Harley, she smiles, but for a different reason...
Meanwhile Zedd's face is always "smiling" making him a perfect stand in for the Joker....except he is smiling for the opposite reason the Joker is: he is in love...
