Well in the KlK comic in the NewType magazine Ryuko did consider calling her "Satsuki-nee" (along with "nee-san", "onee-chan" and "nee-chan"). The magazine was released right before the OVA and I kinda hoped that we get to hear "Satsuki-nee" so I was a bit sad that she just went with "nee-san"... though that was the obvious choice xD
