She's probably sad because everypony keeps calling her "Derpy" because she looks and acts different. I don't think that's really in line with the theme of tolerence and respect in the show.
Personally, I call her Effervescence because it describes one in high spirits (a "bubbly" personality), but also connotates escaping from confinement. She has had to struggle so much in her life, and she has finally overcome and found her happiness in life from her fun job, her loving friends and, of course, her wonderful daughter.

It's pretty clear what happen here. Derpy just looked though all the mail and found that none were for her. She is deeply sadden because today day is her birthday (March 1st) and this is the third year in a row that her family didn't send any mail to her because her derpy eyes make her an outcast in her family.
Don't worry Derpy. Your fans will send you enough heartwarming mail to last you a lifetime.
