Ah, let's see... Omakes, and other humourous series/anime/manga/movies are good inspirations for coming up with something funny. Before you start making something funny though, think of your target audience, first -- is there a specific fandom, or group of people that you intend to present you humour?
Making something funny is actually not that easy, because people have varying sense of humour, like, some would easily laugh at a simple joke, while others don't. There are also some jokes and references that other people might not seem to get (which tends to happen in some of the comics I make, even if it's Bleach)
There are also times that you have to exaggerate a bit of a character's trait/s, but be careful not to make them too OOC. And be careful also of jokes that might be offensive. Some topics that you should be cautious about involves gender, religion, race, and in fandoms, ship and character hate, very serious events (like character deaths, especially of popular characters), and hate on the series and its creator.

Yeah, I usually try to keep them in mind. Though I try to lighten a serious situation (like when Rose and Kensei got beaten by Mask), I still avoid stepping off the boundary and offending people who love a certain character, or even Kubo-sensei himself. It's all about still trying to respect others.
Omakes are those short clips at the end of every Bleach episode, like the Shinigami Cup and Arrancar Encyclopedia.
