The longevity of alicorn is thanks to their regenerative and healing magic. That's why alicorns don't die from old age. They would die from old age only when they would stop regenerate themselves.

Should Celestia die (through say, a epic sacrifice to save all of Equestria), would Twilight take over her role as Equestria's ruler?

It's also a good metaphor for what's happening in the show right now. Whether
whether she likes it or not, she's going to have less and less in common with regular folks. Her princess-type duties are increasing (as seen in today's episode) and when it's your usual friendship problem, the map doesn't let her in on it. To get to Gilda's softer side you need Rainbow, to get the baking squared away you need Pinkie, but a demigod's role is elsewhere. And her home is now this big castle that doesn't feel like home. Her path is away from being that librarian in Ponyville and she'll always treasure her friends and enjoy spending time with them but her life and their lives just aren't the same anymore.

There's no evidence that all the princesses live thousands of years. Just because Celestia and Luna did doesn't mean Cadence and Twilight will...It's never mentioned or even hinted that they will

It's also not mentioned or hinted that they won't and will die soon. All we know is that the only alicorns we know lived that long.
And considering the show will likely not tell us one way or the other, why not let people do whatever they want with the idea? It's not like there's right or wrong in this case. The show is not interested in exploring mortality.

I figured they wouldn't address it in the show, which is why I'm going by what one of the producers said in that tweet - that Twi won't outlive her friends xD And it's not that I have a problem with people doing what they want with the idea. I just don't like the idea of Twilight being immortal and outliving everyone she loves ;d

Sure Twilight outliving her friends would be sad. But so would she dying.
And Celestia having to attend the funeral of of of the few ponies she allowed herself to grow really close to and that was like a daughter to her, and Spike having to attend the funeral of the pony who had been with him and caring for him since he was hatched like an inseparable mom/big sister. Don't tell me that's less sad.
Plus, if Twilight lives on, so do the mane six and their friendship in a sense. At least she'd be able to carry their legacy and memories whereas if she dies too, it's kinda over.
So between two sad scenarios, I'd pick the one that keeps one favorite character and mane six alive rather than none.

Celestia and Luna have probably already had to deal with losing a lot of loved ones, but there's nothing that can be done about that since they're immortal and almost everyone they know isn't. Yeah, it is sad, but they must be used to it. Also, Spike would be sad losing all of his friends, but he's not immortal as far as we know. He will just live a lot longer than his pony friends...He'll have to learn to move on and maybe find friends who have a closer lifespan to his. He could do it, and he wouldn't have to continually live with the pain of losing those close to him forever since he'd eventually die too.Β

AFAIK Alicorns don't live forever either, rather they just age very slowly. So whatever the case none of the four of them would truly be immortal, just very long lived compared to regular ponies.Β
And what I meant is that Celestia apprently isn't close or very attached to that many ponies asside from her sister, possibly because of the unique position she finds herself in. Twilight, as fate would have it, became one of the notable few exceptions to that and is all but a daughter to Celestia, so it would be especially painful to lose her.
Same for Spike. He grew with Twilight and he said it himself that she is his family, all but a big sister/mother to him. We saw that Twi leaving him was his deepest fear in Crystal Empire. So I'd say losing her would be as painful as Twi losing her friends, if not more.

I at first thought that the size difference here was exaggerated, but looking back at screenshots Celestia really is nearly that tall compared to the other ponies. She is considerably more thin and lanky, though.
