It's a line between real and not-real world.
I think you better understand it with an example.
If any character points out something like "Not before all those people!" and points at you, as the one watching them, they broke the fourth wall.
Pinkie Pie from MLP:FiM is a good example - talks to watchers all the time, in one comic she says she can't sing because noone would hear it in this form anyways.
That's basically what Puppet and Springtrap did - they pointed out WE are here and WE are actually reading it. And they looked at where should be just a wall, but where WE are - they looked at people behind the fourth wall.
