I don't have speedpaint. In fact, I'm not really sure what speedpaint is. I know uses Photoshop and Paint Tool Sai. I use Corel Photo Paint. If you download the blank meme (there's a link in the author's comments above) and then you can upload it to Paint and do it that way. Or, there's dA muro.
If you want to use dA muro, scroll all the way to the top of the screen, and you'll see the Submit tab. Hover your cursor over it, and this dropdown box appears. One of the options on the dropdown box is "Draw with dA Muro". Click it, and have fun drawing.

Okay, I know what you mean now. Actually, what they were using was probably some sort of drawing software, while they just used PowerPoint or Windows Movie Maker to make the videos.
