I use two different programs, FireAlpaca and Paint Tool Sai. FireAlpaca causes the background to be transparent.

The background is already transparent. I typically will do my sketching in FireAlpaca then transfer to Sai for polishing.

Save the file and open it up in Paint Tool Sai.

I think JPEG will work but I usually save with a PNG file.

Yes, the checkered grey background will appear as white, but when you upload to DeviantArt it will still appear as trasparent.
