I think you mean RGB vs. CMYK (you can still use layers in CMYK). Or maybe you mean layers vs. channels...
Well I'll say that I prefer RGB to CMYK since i get more color options, even though not all of them are printer friendly. I can see the value of using channels instead of layers for coloring since it reduces file sizes (which is awesome) but that kind of coloring requires you to be braver when rendering.
I guess it all depends on what I'll eventually do with the picture.
