Wouldn't this be a funny idea? Doof and Perry switching jobs for a day.
Perry is now the Mad Scientist and Dr. Doof is now the Secret Agent. Although Doofy is more Johnny English then James Bond.

They really need to make an episode where that happens. I can imagine it Phineas and Ferb are trying to bust Candace and Doofenshmirtz is trying to stop Perry from taking over the Tri-State area.

Their was that one time in "Hail Doofaina" where Vanessa and Candace switched clothes. Even the characters switched lines, like Doof telling Norm "What they're going to do today" and Phineas describing "The Rainboinator" the way Doof does.
