I find several problems wrong with that.

Spike say to Twilight that she's not her mommy. And then twilight says to spike that she is not his mommy so therefore twilight agrees with that fact even she said 'Buck you' to him?'

In the Winter Wrap Up episode (ep 11) Twilight is trying to get Spike to wake up and Spike wakes up saying:
"Mommy?... You're not Mommy" and goes back to sleep. It's a cute joke.
In this comic I added Twilight remembering that she hatched him (ep 23), and since she's more or less his boss, she comes back at him with a phrase that states "Like Heck I'm Not!" or "I MIGHT AS WELL BE"...
I chose "Buck You 'I'm Not Mommy'!" because it's the same style of mocking rebuttal that was used in the MLP spoof "APPLE.MOV" (It's on YouTube. Warning: Some offensive language).

So, in essence, he's correct. At best Twilight is his sister figure.

I Always did think of Twi/Spike as siblings and Celestia as a female role-model/mother.
They never talk about Twilight's parents. We assume that AppleJack's folks passed on since AJ, Big Mac, Granny, and Apple Bloom run the farm. What's Twilight's relationship with her family like? Fun to think about
