While the concept of Human Twilight being a Mad Scientist isn't exactly an original idea, I feel that you do an excellent job of bringing the depiction to life here. First off, the lighting. The rays of light shining down on Human Twilight really help bring out the character's insanity, and I love how you made her sclera (eye whites) a little bit blue to possibly indicate the presence of Midnight Sparkle.
The grin on her face is also well-done. You can see the eagerness on her face as she reaches for a whole new world of magic to explore. And the slightly frazzled hair indicates her increasing loss of sanity. The glowing lights of the background machines and the syringe in her hand only further hammer in her utter detachment from reality.
And then there's Pony Twilight. The sweat on her brow and shrunken irises are only natural for a person stuck in the situation she is, but I feel like the image might come across better if her mouth was open, either screaming or about to scream. But that's just a minor nitpick.
All in all, your work is excellent. And I would eagerly like to see more done with these two.
