Just set the size of your brush to 1 pixel, and I removed the background using Paint.net.

Try zooming in, maybe? On a 50x50 picture you should probably zoom in anyway. Don't draw lines, but try to make lil squares instead.

I wasn't done playing with it. x'3 I changed the colors and fixed his ear and hair, now it's done even if it's not animated.

I don't have PSD, but you could open the PNG/JPEG file on Photoshop and make it PSD.

I should probably make the other frames for the animation too. x3

I think .tiff supports transparency and .sai doesn't, otherwise I'm not sure what makes .tiff special

if you had three seperate png/jpeg files, you could still swoosh them into one file over photoshop
