On the contrary, I believe they didn't change size from the transferring from Drene to Earth, which means they are naturally that size. Drene probably is equal to or bigger than earth, which would mean that their whole body structure would have evolved to handle the increased gravity. Muscle tone, bone strength...

It really wouldn't then. Wouldn't they create a gravity signature about 1/2 the size of the moon? Wouldn't that disrupt the natural balance of earth? Even the moon in Majoras Mask would disrupt the balance so these giantesses would definitely do it. No offense and I know it's all fantasy but I just wanted to point that out

The Drenics are big, but they're not THAT big. Remember the Moon is much bigger than people give it credit for. However, we as Humans have never dealt with a organic form of a Drenic's scale before. Changes the equation, since organic beings can have size but they're not dense enough to create as strong of a gravitational field as say... an asteroid at the same size. Asteroid technically has more mass packed into the same amount of space to create an actual gravitation field.
It's all conceptual, and interesting to talk about. So lets!

Even at the x500 scale, human/drene interaction without technological support would be EXTREMELY difficult. Possible, as a Drene could put a finger next to a ledge, have the Human embark the finger, raise said finger to ear, and the human yell into it... but yes, interaction would be pretty much impossible.
With the assistance of technology though, it's possible, as we saw with the Skype and Phone calls and such... granted the fact they had exact same technology with exact same frequencies says that they may have come from an alternate universe instead of a different part of ours.

But the relationships that they made with the humans, the romantic things we've seen. Those are really not going to work with this scale. Might as well just go back home. I mean seriously, why can they do with the humans now?

In all honesty, there are relationships in real life were people don't have too physically intimate relationships (for whatever reasons). It may sound cheesy, but if they get into any relationships, they'll be genuine "love" relationships. Once they get passed the building sized fingers that would probably pulverize someone nearly instantly... Point being, there are theoretically / conceptual ways around pretty much every aspect of the story so far.

I understand that intimacy isn't necessary for the relationship but for actual organic intelligent interaction that really bond people together I don't think it is really possible. Talking to someone online everyday doesn't really compare to real life interactions. I have some experience myself and I think you may have too. It isn't an uncommon realization

In all honesty, the one time that I did have an online relationship, it almost felt more real, even though I knew that she could've cheated so easily without knowledge. The trust has to be extremely strong for an online relationship to work, and this often gives them more character than personal ones.
On a note back to GatC, I'm not quite sure what you mean by "actual organic intelligent interaction"... If you mean by activities, there are things to do that can bond, depending on which Drene. For example, worship Scarlett, you're good to go. Go with Janine to see a sunken New York, you're go to go... or go for a water park visit in Janine's mouth, you're good to go.
Tbh, the way Organic works (his name is spacing me, so I'll call himΒ X right now) when X was kissing Janine's tongue, that is sharing saliva. Organisms bond much more when saliva is shared, that's why all mammals kiss/lick each other on the mouth.
