Depends on the dosage, effectiveness, and how long the serum remains in his system. THIS time around his mind's still around, though.

For some time, yes. Riley has built up a tolerance to the serum, and it can't naturally produce inside of his body. An overdose would leave him an otter in body and mind for up to several weeks, but the serum would naturally drain from his body overtime.
The real issue come from said draining. An overdosed Riley's bodily fluids would all contain a high concentration of the serum as it is systematically rejected by Riley's body. Coming into contact with these fluids will infect and transform the contactee. And, unlike Riley, who has his built up tolerance, the rest of the victims have never encountered these serums before, and thus it can recreate itself inside them, thus causing a major biological outbreak.
So basically, an overdosed Riley is Typhoid Mary if she was infected with the T-Virus.
