I don't know if anyone's pointed this out, but Leo's eyes are the wrong color. They're blue, not brown.

The artist could have been looking at a reference image from the 2007 film, in that film leonardo's eyes are brown.

Or the artist could've been using that one yes. When I looked at the the eyes from 2003 season 7 it looked more blue than brown, though.

Up until... today, this really bothered me - not your art but the concept. Then the biologist in me jumped all over my "human hang-ups". Nature is all about reproductive drive: if you can't love the one you want, you love the one you're with. So, the boys would have literally no one else to turn to and a distinct lack of human influence (or anyone's really, cause Splinter? he's got no clue about turtle selection) telling them that they "shouldn't" do what comes naturally. Furthermore, there is nothing to tell us that they actually come from the same hatching, let alone the same parents. Now, Donnie's cannon crush on April is much more unnatural because it goes against nature; they are biologically so far removed from each other (forget species, they're not even the same Class) it's doubtful either one would even think of romance or sex. Thus, I now get to fully enjoy you're lovely artwork.
So! I love their expressions. Each one of the boys is thinking the exact right thing. Donnie already knows what I just figured out, he just would like you to leave, we're busy. Mikey doesn't even get that what they're doing might be weird for someone else, so hello. Leo's protective and assessing the intruder, because they're fine but other people can be really stupid. And Raph's a bit wrong footed because he was just caught completely off guard; yikes! when did you get here?
I totally dig the in-a-church-made-of-stained-glass vibe that's going on here, but the rose and the vase just don't fit in. Maybe if the vase was crystal and casting shafts of light instead of these bizarrely long-stemmed, moderately battered flowers. They are done well, but they look like they were plucked from a different picture and just dropped here. I agree that the table needed something, but I think this choice was mismatched. Am I beating a dead horse? I don't think so, but I'll stop now anyway.
I also really appreciate how what they're doing is so obvious without being totally lewd. Not that lewd pictures aren't great, but they're TURTLES. Teenage Mutant Ninja Anatomy Lesson very much not needed. This is PERFECT. Sensual and caring and almost human, because so much about them is Almost-Human.
The luminous quality of Mikey's eyes might just be my favorite part of the whole thing. Raph's are pretty awesome too. And the body language, all of it.
