In the film mewtwo in born from a 'test tube',
Mewtwo was cloned from mew and spliced with a lugia and given his own genes by the humans that created him.
Mew is considered unique,but I suppose not nececarily is, Though Mew is seen as a sort of 'god' of the pokemon world.

The way I understand it, originally there was more than one, but there's only one left now. The fossilized eyelash could easily have come from a different Mew than our lone survivor.

You're right about Mewtwo's genetic base having come from the eyelash of a Mew which would logically now be long-dead. But I don't think there's any reason to believe that the Mew seen in the film is the only one. What about the Mew from "Lucario and the Mystery of Mew"?
Think about it: the Mew in "Mewtwo Strikes Back" has met and interacted with Team Rocket, Ash/Satoshi, and Pikachu. If that Mew was the one later seen in LatMoM, why does is "abduct" Pikachu and Meowth and try to keep them from returning to their human companions? The Mew in MSB was on the side of the humans and their Pokèmon, and would understand the bond between trainers and Pokèmon; it wouldn't try to keep Pikachu from going back to Ash after having seen what a strong friendship they share. If anything, it probably would have revealed itself to Ash and tried to play with him, rather than resorting to "stealing" Pikachu as a companion.
So, based solely on the movies, I think it's likely that there are at least two Mews in existence.

That makes sense. Their behaviors aren't that different; Mew is secretive and prefers not to show itself, but playful and benevolent. However, I can see your points, and I'll add one of my own; the Mew that Mewtwo knows is strongly associated with water, specifically Mt. Quena (from Mewtwo Returns), suggesting that it lives there. The one from Mystery of Mew obviously lives in the BIG STONE TREE that was the FOCUS OF THE WHOLE MOVIE. If you really want to go an extra length, observe that the place where they found the eyelash (and where we, the audience, saw a living Mew) appeared to be a rainforest of some kind; not the kind of environment likely to be anywhere near either of the first two. One could argue that this is evidence of at least three living Mews.

Ah, good point. I haven't watched the first movie in a while, so I wasn't sure if we see one Mew or two in it.
And while there's no reason to assume that Mew couldn't up and move if it wanted to, it's made fairly obvious that the Mew which lives in the Tree of Life has been there for hundreds of years, since we see it in the flashbacks of the "medieval" Pokè-world, where it assists Lucario's trainer.
My personal feeling is that there's a breeding population of Mews, large enough to be sustainable, but small enough that they don't normally come into contact with humans. (Of course, given Mew's Transform ability, there could actually be lots of them, venturing into human cities and even living side-by-side with people in the guise of more common Pokèmon.)
