Doomguy: Is only wearing security armor, and only on the upper half of his body.
MasterChief: Beat 4 highly trained marines to death while boxing with them, when he was fourteen, before he even received his armor.
Samus: Isn't even IN her armor half the time, and has kicked ass in this state.

But I don't know a lot about Freeman or Samus since I don't play their games.
But I think it's safe to say that Duke Nukem has killed the most lol.

Doomguy has killed the armies of hell on 5 different occasions.
Masterchief has defeated entire armadas of coveneant soldiers and entire battle cruisers and PLANETS by himself
Samus has done pretty much the same, and giant monsters
and Gordon has done something similar to a lesser extent

Duke Nukem has committed Xenoside twice, killed his nemesis and his whole army twice and has gone on to shoot more bullets than thought possible.
Doomguy has killed a lot alright, there's no argument there. I'd say Doomguy and Duke would be about evenly matched.
Master Chief has killed a few thousand Coveneant and other monsters but not enough to match Duke, he hasn't even disbanded the Coveneant yet.
Don't know much about Samus or Gordon but I doubt they've killed as much as Duke.

Chief may be on par with the marine from Doom and Duke, but it really depends on how much of the Flood and Covenant were left on the two Halo installations and the Ark, which is unknown.
But two accounts of xenocide might alone be more than that anyway.

Depending on which enemies you count.
I'm counting sentinels, flood and all affiliates of the covenant, which would put his kill count far above that of the other two....depending on how many soldiers the armies of hell had.

Can't forget the Sentinels and the Prometheans...especially the Sentinels since there were billions of them working on the Halos and trillions of them on the Ark.

Master Chief has been fighting in the covenant war ALONE for 27 years, killing upwards of 10's of thousands of covenant during the course of his normal duties.
Before that he was even fighting rebels for an a while, and who knows how many people he killed there.
Then we have the ACTUAL GAMES where he DESTROYED the entire halo ring, along with the MILLIONS of sentinels and thousands upon thousands of covenant and flood.
Then the sequel! Where he destroyed high charity and killed every covenant there, and not taking into account the large amount of covenant civilians which would also number in the millions for the capitol city of the entire covenant, he still killed thousands and thousands of soldiers.
Then halo 3! where he destroyed the thing that fires all of the halo rings and killed thousands upon thousand upon THOUSANDS more covenant millions of sentinels and oh so many space ships with even more sentinels, also destroying the gravemind and every last trace of flood there is, which could be any number really, maybe even millions by itself.
Then in halo 4, where he didn't kill very many enemies, but he did kill the modern equivalent to a GOD. And flew a sick-ass starfighter into the deathstar to do it, all the while still making his kill count go up by killing Promethean and covenant.
Oh wait, they're not even covenant anymore, as that was disbanded.
These are straggling space pirates who still call themselves "covenant"
the actual organization was disbanded due to lack of leadership after the chief whooped their prophet's asses.
Duke cant compete with a man who has literally made something EXTINCT. And destroyed entire ARMIES of multiple races sizes and strengths.
All while carrying only two guns.

So Chief making 1 alien species "near" extinct is better than Duke Nukem making 2 entire alien species completely extinct?
Duke 1: He fought his nemesis and his cyborg army, killing all of them. Using only 1 gun too. He even traveled through time.
Duke 2: He fought an entire alien civilization on their home planet(again with 1 gun) and made them extinct.
Duke 3D: He fought a 2nd alien civilization and ended up destroying all of LA while killing off every one of the aliens on earth sending whatever aliens that remained to run off and find their allies for help. He had a lot of cool guns then too.
Time to Kill: He fought the same civilization that allied with a lizard civilization(can't remember their name) again through time, in the present and even back to the old west, to the mid-evil times and then the Roman times. He Killed them all. He did all that again in Zero Hour, while fighting zombies and even killed Jack the Ripper lol.
Land of the Babes: Fought and Killed another allied civilization that wiped out all male human population on earth, he killed millions of aliens and repopulated the entire planet earth by himself.
Manhattan Project: Long story short, he killed another allied civilization that not only allied with a lizard army, but also a cyborg army.
Forever: Finally with no more allied alien civilizations left to kill, Duke Nukem wiped out the entire Cycloid civilization Empire and their Emperor. Straight after that he was kidnapped by his nemesis from the first game and his new army of Duke Nukem clones. Duke killed Proton and the thousand copies of himself and then killed the Cycloid Empress.
Duke did all of that by himself with no power armor and no help. Just him and his guns.

Almost none of that can even be considered factual as he just killed a bunch of Aliens and assumed them extinct.
Also you can hardly say he did it by himself, seeing as he would be DEAD if it wasn't for that helicopter rescuing him Forever.
ALSO gunpowerups and general powerups and inventory items are the same if not worse then power armor and having a nagging house wife in your head telling you to go to places you already know to go.

Well his most recent title was stuck in hell for over 12 years so it's understandable. However his next game will be awesome. I think it's likely Duke Nukem 5 is going to be a Next Gen game and it will either be announced or released on his 25th Anniversary, which is 3 years away. If you think about it, that is very likely because Gearbox needs to get Furious 4 and Brothers in Arms done to fix up their rep after what happened to Aliens, that would take about a year in a half since those games would be about half done and then GBX can take another year in a half to make Duke 5.

Gearbox did well with some Half Life expansions and such, but yeah they are very lucky Borderlands worked out, I just hope they put the same amount of effort that made BL2 an award winning game into Duke Nukem 5.
