Not really, since he was hatched by her magic. Twilight didn't lay the egg, so Spike isn't blood related to her... Unless magic has that kind of ability. I don't really know how incest works that much and I'm lazy so uhh I'm not a reliable source.
But it would still be weird because Spike is a teenager and Twilight is a full-grown mare (let's not get into EQG) and takes care of him as if he were her own son.. kinda sort of.
