Here's the thing, though; Spike doesn't have to die. He just has to be emotionally hurt enough by Twilight prioritizing her books over him to not want to be her assistant anymore. Then Twilight would not have both

Hurt enough? How would he be hurt? He's immune to fire and he fully knows it, right? So he could wait for his turn, while trying to put out the fire.
Come to think of it, dragons would make great team members for fire departments.

If Twilight decides to choose saving books before Spike, it's a sign that she considers books to be more important than Spike. If a close friend of yours decided that a collection of books meant more to him than you, wouldn't you feel a little insulted?
Here's another thing: have you ever been inside a building that's on fire and could possibly collapse at any moment? With no way for you to get out? I am pretty sure it is not a pleasant situation. And Spike is still just a child; even with fire immunity, he would still probably be scared out of his mind. Being a dragon may make you immune to fire but it doesn't make you immune to fear itself. Imagine Twilight scrambling to save some books and scrolls over Spike's panicked yells and wails. That's still pretty cold, even with fire immunity.
And as others have pointed out before me, Spike can still be harmed by the building collapsing down upon him. Spike could still die by being buried by debris and suffocating

Cold? Fear of the fire? Even though Spike casually swam in a volcano's lava lake, without a trace of fear?
I don't think so.
Besides, even as a kid, Spike is more than old and mature enough to know that if he swims in lava, he has nothing to fear from a house fire that has just started.
And if the fire has just started, there are no risk (at least, not yet) of falling debris or collapsing of the building.
Now, how long would it take before the structural integrity of the building is compromised?
I'm not sure. Probably not long enough to save the whole library, since the entire building is a tree.
Really, the Equestrian national and local authorities should put much more effort in fire prevention and rescue services.

The thing is, we don't know if the fire had just started. We only know that the the library is currently on fire, not how much time has passed since the fire started. So there's still the possibility of Spike getting buried under debris and suffocating. And, that being said, there's still a reason for Spike to be scared and go panicking. As well, Discord has made it clear that Twilight only has time to save one item. So I think the implication of what will happen to the item or dragon she didn't save is pretty clear.
I'd like to re-emphasize my original point as well: Even if Spike doesn't perish in the burning library, if he ends up feeling betrayed or emotionally hurt by Twilight's decision to the point where he doesn't want to be with Twilight anymore, Twilight will still lose Spike. Look at Spike's reaction in the comic. It doesn't look like Spike is happy with Twilight's choice. And if this is his reaction to Twilight hypothesizing, how well do you think he'll take it if Twilight acted upon this decision in reality?

But if she saved Spike she would tell him he have to rewrite all the List, and all the Research over again then he relized she should have picked the research.
Spike should think who does most of all the writing for Twilight.

So unless the whole tree-house is crumbling down, Spike can wait. And make himself usefull by trying to put out the fire, rather than freaking out in front of a element he's totally immune to, anyway.
