Rainbow Dash's first thought wasn't that far off. They are both reptils, have both scales and their two races are at least distinctly related for sure.
It was only later on when she went a bit over board with saying "You're practically twins!", but her guess that Spike could know something about hibernation was a logical conclusion.
And Spike was too sensitive about this.
