Almost a year went by since my last Tutorial...Sorry for that!!Well remeber the poll I started months ago? Well a lot of things got asked so i decided to do a stap by step again, recording each step I do.
I already did it a few weeks ago but just finished it a few minutes ago. The final illustration will be up on my Tumblr soon (and a bit changed).

i have two questions, if you dont mind
1. when you selected your character and it turned grey, did you do make it that colour on your own, or does it automatically become like that?
2. what does the multiply tool do?

This answer is for anyone reading this in 2016 or later who is curious about the same question: here is what "multiply" does: imagine that you have two differently colored pieces of transparent plastic, and you put them on top of each other. The effect is that a new color results. The "multiply" setting turns a layer into such a transparent piece of plastic, so that it is influenced by the color of the layer beneath it. In contrast, "normal" acts like you had two completely solid pieces of plastic: so if you put one on top of the other, you'll only see the solid color of the layer on top.

Yes I filled it in with grey by myself. So paint tool sai is not doing it automatically.
It's hard for me to explain, it's a setting/effect fo the layer not a tool. When you do a new layer above a already excisting one you can draw over the very first one and it turns out darker, you can shade this way without losing the details below.
Argh I cannot explain it I'm really bad with words or describing, please try it out and you'll see what I am talking about.

Do you know a trick how the "Magic Wand"-Tool work again? My don't want to work :/ Even if there is no gab in the line art, it marks everything! ._.

If you are marking everything, then either:
1. You are not using the magic wand in the same layer as your line art; or:
2. You have a gap/opening somewhere in your line art that you haven't found

Ah yes you need to select the area around your character (the bg) and that invert it. than your character/lineart is selected.

I just fill it in so I have basic color, but the actual reason is that I have it filled in and can select the character whenever I want. This saves time and I won't draw over the lines
