Correct me if I'm wrong, but from what I saw, Gumball wasn't responsible for the way how Rob turned out; it was an accident of Rob's part.

well I mean, it seemed like they were sort of making it like data corruption, so it would be a matter of restoring the broken code. Dunno what that would mean in-universe though
lmao I have a flashdrive tho that is mostly Rob and AWOG stuff and I accidentally corrupted it so every episode on it is basically The Signal in level of glitchyness but still watchable

Why was Rob a mistake? I don't think the show ever explained why.
I think Rob only hates Gumball he associates his "glitched out" form and how it got that way because Gumball was around when it happened, and Rob was such in a panic that he couldn't think straight, so he just blames Gumball for it. Also, Rob had different way of thinking before this transformation. So, maybe the distortion has messed up his state of mind? Could be.

Well, they used the word "mistake" kind of loosely. They meant it less as an "error" and more as something extraneous and unnecessary. A character who's main personality trait is being boring, or one who's main trait is getting incredibly offended when someone gets his name wrong, are not characters who are *coughs loudly* likely to get deeply involved with any sort of plot line or likely to be remembered. Though the show never really explained the specifics of how or why, there was an implication that the void was more for things/people that got discarded for one reason or another.
Also Gumball is kind of a complete asshole to him even though Rob is kind of an asshole back so it's not like it's only that association that created that antagonism.
I think it's less the distortion that's messed with Rob's mind and more the reality trying to throw him away and erasing his memories and forcing him to hide in someone's house for months and finding out his entire universe is just a cartoon people in another universe watch to entertain themselves and the frustrated sort of dissociation that comes with that...

well, yes, but Gumball also has a bit of protagonist syndrome in believing that the world pretty much revolves around him because it sort of does, which makes him overall a bit less sympathetic to everyone else. He'll shove himself into other people's lives because he needs to be involved in everything and he wants to have a nemesis because it makes him feel more important and makes his life more exciting even though most people really wouldn't want someone around who desperately wants them to not be alive.
