I don't think you understand that quote completely. Mark Twain hated politics altogether.
He also didn't believe that politics mattered as you can see in one of his quotes "if voting made a difference, they wouldn’t let us do it"

I can understand why he felt that way he grew up under the democratic south where slavery was not only legal, but celebrated and then he lived to see the long reign of the republican party where wealth was valued above all else. His quote is not so much about hatred of the republican party as a joke about how voters never trusted anything, but republicans to be in office.

I mean he is the man who once performed a mock funeral of the democratic party for the republican administration of the time.
