When Karai became a Vampire (Vam Mi attacked her) she decided she wouldn't drink human blood, which spared her Foot soldiers. But she still needed blood, and so captured Leonardo, conscripting him into servicing this need. He did not agree to it at all.

Will he become a vampire too? Will they fall in love?

They may fall in love in this one - I'm not really sure yet. (In truth, the emphasis of my other comic is the romance between Karai & Leonardo.)
But canonically speaking, according to the '97 series, the turtles can't become vampires because they're not human.
BUT, part of them IS human, so that part can definitely become enslaved by a vampire.
