I have been a Spyro fan since the games came out for PS1.  I also enjoy MLP: FIM, especially Spike's character.  There is a bunch of overlap between Spike and Spyro, so it makes great comedic value for a comic.  My original plan was to have the comic be Spike and Spyro: College Roommates.  It would have portrayed Spike and Spyro as undergrads dealing with typical college shenanigans.  I then wanted to have the mane six (Twilight, Rainbow Dash, etc) be their TA's and RA's.  This comic evolved from the idea of Spike and Spyro being freshman in a entry level college class with Twilight as their instructor.  I also threw in some influence from Evil Dead: Army of Darkness and Skyrim.

Yeah, I'm a big Spyro fan too and have the same fascination with Spike meeting Spyro.
Spyro is the spunky little dragon hero with attitude, wonder what he'd say meeting Spike for the first time, because Spike's a little effeminate from spending his entire life around mares.

I've actually been working on a comic of them meeting for the first time.  I've been doing much more detailed vector work this time though, so it takes quite a bit more time.  It should be pretty good though once it's finished!
