cca 45x125x enlarged , H&E, taken with Carl Zeiss Jenaval.Let's have another histology. It's undecalcified coronal slice of mouse nose this time - it belonged to male of Nu/Nu nude strain. It's quite thick slide, I'm supposed to do better work next time ;]
Anyway, enjoy nice nose cartilage. Also Nu/Nu is nude strain but isn't free of fur at all , especially on nose - note plenty of hair follicles and bigger whisker follicles.
Backstage story - at times in which was this slide made we had problems with rats in lab. Our den is in old cover which was empty for some time so those critters was used have refugium there at cold winter. We thought alot about printing of this pic in A0 format as warning for every mouse-like creature with tendency put it's curious nose in places where they aren't allowed anymore ;]

Very nice! How thick is it? And how did you stain it? Embedded in Paraffin? I have never seen a whole head prepared, only soft organs, so I am curious on how it works. Do you have a special blade?

This one is stained with basic protocol where hematoxylin and eosin are combined. I prepared only tip of nose and organs, so no decalcification was needed there, it fitted in normal workflow then. And yep, after processing in alcohol and clearing agents it continues to paraffin blocks. Microtome was set to 10 um, but this is sure more thick layer. Same thickness on every slide require tons of skill with preparation , there is 1000 ways to screw it because protocol is quite long.

Meet my Meopta microtome - it's vintage machine from 1967 www.sumie-dh.cz/images/gallery… . You can see blade there, I have four of them - most compliated thing was finding of cutler which is brave enough to sharp them without damage. As removable razors are now used in modern microtomes, people with those skills slowly vanishes.
Also, note that adult mouse is too big for standard 76x26 mm slides. Even few days old mouse must be spliced to parts. There is also big problem with tissue fixation, it must be done by putting fixative solution directly to veins of narcotized animal with method called perfusion. That's something behind my skills, ethic will and courage, those methods are supposed to hands with veterinary skills. This male was only putted down with ether and then dissected.
Ad decalcification - we use mix of sodium citrate, formic acid and water to get rid of unwanted calcium.

Thank you for the insight! It has been quite a while since my time in the histology lab, so this was a nice refresher!
Nose tips are not prepared in our labs, since it is veterinary histopathology and mostly focuses on ruminants and carnivores (dogs/foxes). So I find this preparation especially interesting, although I am very unfamiliar with it.
I guess my anatomy teacher would have loved to have a picture like this for her lessons. The structure of the nose and the sinuses was one of her favorite topics. She even had a skull cut in half to proudly show us the fragile walls inside (but don't you even think of touching them lol)

PS: When our freezing microtome broke, we got the old one out of the basement. It was similar to yours in age, but the sounds the freezing apparatus made were horrifying at times xD
I understand your struggle with the thickness, the old guys sometimes need a little pleading
