So, I don't know if this is what anime looks like to you, but this is what anime looks like to me.Ideally for Illustrator CS2 users but these principles/tips/techniques can be applied to pretty much any program.
This is my contribution to Vector Week; July 21-25.

"Triangle Cheek Lighting" as you call it actually already has a name. It's called Rembrandt lighting, named after the great painter who so often used it in his work.

My lineart is composed of shapes that can be filled, they are not created with the line tool but with the fill tool that is part of the pen tool.

I usually keep all my colors, including gradients, on one layer. I have the lineart in another layer above everything. But that's really up to you.. You can make as many layers as you need for each gradient or set or gradients.
