UPDATED 10/31/2011: Made a few enhancements based on feedback below. This involved fixing her cutie mark, stiffening her hooves, fixing the right hind leg, adding an overlay over her horn, and improving the blink animation.The source file has been updated below, but the original source and gif can still be downloaded here:

My most ambitious test of Synfig Studio yet, featuring Twilight Sparkle in her regular walking animation.
It took a while to do only because I was still getting used to using this program for something more complicated than stick figures, but once I got the ball rolling it was fairly easy-going. [although it still needs more polish, of course] ^^
All the animation was done using only 5 waypoints (6, if you count the blink), with the hair and cutie mark bobbing being handled by Warp Distort layers. The additional 5 waypoints needed to include the blink animation were added by duplicating the original 5 waypoints -- note that the Time Loop layer is only there to create the Youtube video rendering, but is not needed for the regular GIF render.
Also, since these characters aren't mine, the source is free for anyone in the community to use if they want...

As for how I do these animations....
Well that depends on your choice of software. If you prefer stuff like Photoshop, Illustrator and Flash, I probably can't help you much since I don't use any of them.
[with the exception of Photoshop Touch on Android, but that's a different story]
However the general concepts are still the same. ^^
In the case of the above, I use a combination of Inkscape to create the original vector image, and Synfig to do the animation. But are free and powerful tools -- and although Synfig is still in active development, Inkscape is very mature and easy to use. ^^
If you know about vector art, you'll know that drawing them is fairly easy (more sculpture than drawing actually) -- and likewise animating them is easy too since most of the tweening is done for you after you establish some keyframes.
But if you don't know much about vector art, I recommend starting with drawing still images first in Inkscape. It's a little more fun and easier to use than Illustrator, and more dedicated than SAI or Photoshop's vector tools.
In my description I have a copy of Twilight's vector source for you to look at, if it helps.

I meant that it pales in comparison to the ones I made later, since this one was my first and has a few bugs in it still. On the other hand, it does have Twilight... and because Twi is loved by ALL, it's actually been the most popular thus far.
[least popular was Star Sparkle,... guess people don't quite love Twi's mom as much.... >>; ]
