Most likely the latter, but either way; it doesn't make a whole lot of sense that Twilight would be immortal. Celestia and Luna are obviously immortal, but that is because they are not JUST alicorns. I mean, an alicorn, basically, is just a unicorn with wings (or a pegasus with a horn, if you will). Celestia and Luna are those, but the fact that they are pretty much responsible for a lot in Equestria; they look after the entire land, and raise and lower the sun and moon; they have that flowy mane thing, while Twilight and Cadence don't have that...
I mean, Celestia and Luna are pretty much demigods, not just alicorns. Twilight and Cadence are just alicorns, though, as evidenced by the lack of the flowy mane, the fact that they weren't BORN as demigods, and their inferior magic skills. It just speaks against the whole 'immortality' thing.

Ok, first, prior to their reign, the sun and moon were raised and lowered by the unicorns, so you cannot conclude that they are tied to them.
Second, their ability to raise the sun and moon does not automatically give them immortality.
Third, age spells have been shown to exist, so they need not be immortal to have lived long as they have.
And fourth, there's no evidence that they're demigods, nor is there any evidence that they were born alicorns. That's just your headcanon, and you should never use a headcanon as an argument. And no, their flowing manes does not count, because all that conveys to us is that their manes are magical.

Well, if age spells exist, and Celestia and Luna somehow have used them to stay young, then it must mean they cannot be immortal, because then they wouldn't need an age spell to keep them young for them.
And... The sun and moon were raised and lowered by the unicorns? OK, if that is true, then how come only TWO ponies got that as their talent; how come it didn't happen earlier, and HOW did Celestia and Luna achieve alicorn status if they weren't already alicorns? Like, did the unicorns just rule over themselves, until a couple of random unicorns somehow achieved alicorn status for no reason? Like, Twilight and Cadence became alicorns because they reached some sort of nirvana by committing an act of extreme love and friendship, so how do you reach nirvana by becoming great at raising celestial bodies? That's just something you do, not achieve.
And the flowing manes; why would they have them, exactly? I mean, they don't serve any practical use, so I doubt they are being willingly manipulated by magic from the princesses.

I never said that they did use them, I only said that it is a possibility. All we know is that either they are immortal or they use age spells.

Yes; it's all conjecture! I'm not saying it isn't! At least I've tried to explain my thinking by looking at details in the show, which is something I don't think you were trying to do; all you said was "it could be...", which is true, but if there is reason for doubt in your mind about all I've said, then why don't you give me your own theory (don't bother, because I won't read it)?

...Sorry, but I don't really like this comic. It's way too mean-spirited for me. Which is honestly a problem I've been having with a lot of your comics lately. I simply cannot believe that the other princesses would find these "pranks" funny or think that Twilight would find them funny.
