Two ways:

When I put down the first color (usually a midtone) I zoom in really close and stay nice and neat. I use the eraser if I go over the line, and just generally clean it up as I go. I put everything on a different layer under the lineart, and I hit the "Preserve transparency" button. Since I've got a single, solid layer of say, skin tone, then I can paint the dimensions on top of that without it ever getting messy outside of the lines.

Second way:

Lay out everything on individual layers, paint quick and messy, and spend 5 hours with the eraser tool cleaning everything up.

Whichever way you use depends on what you're painting. Also, certain painterly effects will require one method over another.

As for the tools, I generally use the oil pastel tools on nice and smeary, texture-y settings, and then smooth out the colors with a blending tool. That way I can blend the colors smoothly and quickly and still have some textures left over when its done.
