I would say shortly after the second kiss, offscreen. The fact she had him kiss her a third time implies that she has some kind of third dragon-pony form that we haven't seen yet, and that she told him about it to help his research. When exactly we'll see this third form depends on where the story goes, though it'll probably be a while.

Actually, I read the next page and found out I was wrong. x_x Apparently the real reason for the third kiss was so that Dawn could have a card of himself, as he didn't have one yet. Rarity even said Sparity only had two forms, and I'm going to take that as "word of mom". Oh well! It's fun theorizing, but one could always be wrong. Nobody's perfect!
