It's good to see Spike and Shining Armor having some guy time...or at least sibling time among girls. 
Sweetie Belle's hilarious antics are more than just adorable; it also looks like she's helping Luna loosen up so the princess's funner, wilder side come out and develop a little.
T'is a very nice beach pic with a fantastic ensemble of character! Most folks would only care about including the Element Bearers and the CMC.

Judging by the absence of a cutiemark on that pony that kicked the ball, I'd guess that's SweetieBelle? It's kinda hard to tell from the hair just because it's usually not messy like when she's in action.
WHo's that one that Spike is standing on?
Is that Rainbow Dash that Scootaloo is standing on? Again, the hair makes it hard to tell when there's no color.

Or are the royals not supposed to take part in the fun and laughter? Luna is best princess so she.has fun with her subjects.
Now I wonder. Would.spike consider twi as a big sister or as his master? Spike didn't even get a human role in the movie, which ticked me off. It really shows that dragons, no matter how.smart or helpful, are nothing but less than equal to ponies.

As far a I know, Celestia took care of baby Spike, so he and Twi probably spend a lot of time together and he would surely see her as his sister. I would not put too much emphasis into the magic mirror transformation thing.
When Spike spent time with Twi he probably helped her for fun now and them, which later turned into him being called "Twilights Assistant" - a nickname-like title he felt proud of and kept it up (the title as well as the activity)Β - that's how I think of it at least
