In a fan-comic, Twilight contemplated the answer to life, the universe, and everything with respect to her cutie mark, and essentially deduced that friendship was the translation of the answer.

Also a reference to hitchikers guide to the galaxy 42 is the ultimate answer to everything, and apparently on twilight's cutie mark there are 42 points. Bleh but ya see.
