New and improved!All of the color guides are getting a revamp. The guides are all now two pages, one for digital applications (such as vectors, painting in Photoshop, 3D modeling, etc) and the other for print applications (customs, merch, T-shirts, et cetera). The PNG preview for the digital guide should be OK to use as well, so the PDF is optional.
If you use Google Chrome or Safari, they will attempt to open the file in-line with Google Docs PDF viewer. DO NOT EYEDROP THOSE COLORS. SAVE THE PDF FILE. The Google Docs viewer displays in a very low bit depth that results in dithering of the colors. Until Deviantart gives me a way to disable this, you should use the download file link to view the guide!
They are also now more space efficient, and will be able to scale better with ponies that have more colors than normal.
The Digital guides aim to be as close to the show as possible, while the print guides aim to be as close to official merch as possible. When there's no merch of that pony (e.g. background ponies) then the print colors will be based on my conversions which will aim to reproduce the show without looking too weird or have obvious gamut problems.
It is safe to eyedrop the digital colors or use the hex/RGB codes - the swatches should match up just fine. Just make sure you're working in sRGB color mode.
For the merch guides, the spot colors are rendered using CMYK alternate color space. This is based on my own examinations of the merch boxes, where the spot colors are simulated straight to process anyway.
A note about Derpy: As one of the background ponies, her colors were designed primarily with TV in mind. Some of her colors may not look right, and in absentia of something used in real merch, I decided to go with straight CMYK conversions.

Thanks to these codes, I can convert them into Federal Standard (the colour standard code for minatures and model painting), then paint a show-accurate Derpy Hooves!

Cool, but one small thing is why does Derpy have random red in some of her strokes. I reloaded this tab to see if it was a loading error, but it may just be mozilla.

if you're looking at the PNG preview it's probably because CS6's export does dumber things now (art vs text optimized).

I'm having a real struggle now, because I've been comparing this to my digital copy of Derpy from The Last Roundup, and this looks much bluer. Is this because her color is different from one episode to the next, or is my copy color-corrected incorrectly, or what... I'm not sure what to do about it.

This guide is optimized for a wider range of uses than just the show. It was also made before The Last Roundup. When the updated version of the guide comes out it will have repicked colors optimized for multiple users instead of a compromise biased towards print.
Derpy isn't exactly a neutral gray, she is kind of a cool gray. But I haven't looked at this one in a while.

Okay, that's good to hear. I did notice that some of the others seemed a little more updated than this one. I'm trying a vector, so of course the specific digital color is what I'm looking for... anyway, thanks for your response!

I gotta say, that while I respect your color picking process, and pretty much all the colours are spot on, the blueish gray of the body stroke just simply looks a bit off to me.

I refer you to Sonic Rainboom at 14:54, where I based Derpy's palette from. You've gone a bit too green in yours. Derpy is not very neutral; she's very blue-gray overall. Very metallic.

I agree with your statement about my stroke being too neutral, and the Sonic Rainboom source is a really good one, however even so the SR Derpy seems to have a much darker stroke, or at least a noticeable difference in colour. Maybe I just saw a bad source but here's how it looks like to me sandwiched between your colour guide stroke and my wrong one for comparison: [link]
The reason I thought my version was ok initially, is because colour picking some other shades in my source image led to very close results to yours, exception being the stroke colour. Any insight on this would be appreciated.

Well, it depends on your source. If you check other areas where Derpy shows up, she still has a steel-blue gray stroke. Most of the times when things are very small (as most ponies are in the show) the compression used for HDTV will distort the purity of the colors. It also depends on how you're viewing the colors. If you're picking named colors, the alternate color definition put into the application will make a difference... It's clear I can't please everyone all the time. :V What application are you using and how are you choosing your colors?
Derpy's closeup in Rainboom means that compression will not factor in, for what it's worth. This goes into a really long story that I've addressed in some journal posts about alternate color spaces and things like that.

Thanks for taking the time to explain. I checked your suggested source and it pretty much matched this image in the Wiki gallery(I didn't want to use a screengrab in order to distort the colours less) [link]
As for the method of colour picking, I just eyedropped in Photoshop. To be honest your colour is one of the best fitting so it's not a matter of pleasing me, I was just wondering why eyedropping that image made the stroke look so dark in comparison to yours.

I'm not sure if you saw the other comment I left on our discussion on my profile page, but aside from me having to create some extra fills due to Photoshop's lackluster path export facility the stuff you supplied worked very well. I'd like to do the same thing for a Sweetie Belle guide, if that's possible.
