Alrighty...you've got the skill to draw such heart-wrenching expressions. The 3rd to last panel is among the saddest panels I've ever seen.

It's lucky for those bullies that you're going to be saved by Sunstripe. If Dash saved you, I expect you'd mention these feelings, and then she'd have some significant anger to work off, and a shortage of sensible targets. Really, that scenario just does not end well for anypony.

It may be just me relating to Fluttershy so much, but I don't think anything needs to really 'happen' for her to get like that. Some children are insecure on their own, and thus likely to get picked on from bullies like those two.
And what other kids would just put up with, or react to, makes those insecure kids just more insecure. To the point where they can just not judge their own abilities positively at all.
And then, knowing you're 'bad' at flying discurages you from doing it, thus you never practice, thus you never learn and get even more convinced you can't do it, even though you litterally have to do it all day, which causes the fear.
And fear is a bully much worse than anypony - or anyone - can be to you, because it litterally makes you unable to even think about trying. Which you actually captured quite well in this page, by the way.
There is no one event that makes you like this, it's just a million little things slowly setting you into a downwards spiral. Quite litterally, in this case.
Although if you want to pin her insecurity on one specific, traumatizing event you can do a comic about, I am not going to complain about it.

Although, from the looks on their face (and I'm NOT excusing their behavior here, mind), I"M thinking that the first meanie thought that Fluttershy would just fly when she threw her OFF of that cloud. The "uh-oh" from her kinda clued me into that one.
