I think that it's like said, about The Hitchhiker's Guide to the Galaxy whole thing about the meaning of life being 42 and such, but also on her cutie mark, there are 42 points to all her stars. My theory: There are 42 points to her cutie mark and her cutie mark is about magic. 42 is the meaning of life, and magic = friendship. Friendship is the meaning of life!

Yeah, I know. But I believe that whoever decides to use that number knows it came from Hitchhiker's Guide to the Galaxy. 42 is just a number and can't be copyrighted, but people reuse it randomly as like a secret joke.

I'd say it's highly likely that they gave her that number in reference to that movie/book in the Running of the Leaves
After all, it is THE answer to life the universe AND everything! Definitely suits Twilight.
(Also after a quick google search, apparently the number of points on all the stars on her cutie mark combined is equal to 42 aswell)

Best ones are Pinkie being pi (a delightful pun and in the words of the Joker "What were you expecting from me? A round number?") and the Doctor being "10" (because that's what specific Doctor he is).
